研究方向:

大环对靶点验证与药物的发现起着重要作用,然而,如何设计多样性类天然大环去加快先导化合物的发现与成药性优化具有挑战。我们提出了仿生模块化设计与分子编辑合成类天然大环的策略,实现了合成方法学、化学生物学与药物化学的有效交叉融合,建立了聚焦型的类天然大环(1500余个)化合物库。通过表型筛选,到目前为止,1)成功获得了活性高且毒性低的P-gp调节肿瘤多药耐药性大环抑制剂;2)开发了抗炎活性的非甾体呋喃大环内酰胺;3)发现了抗流感病毒H1N1大环肟抑制剂。
